Abstract Diploma Thesis

The thermosome-encoding gene of the hyperthermophilic archaeal methanogen Methanococcus jannaschii was cloned and expressed in Escherichia coli. The thermosome was expressed in soluble form and the over-expressed subunit assembled into its hexadecameric conformation in the mesophilic host. The thermosome is an oligomer with a calculated molecular mass of 940.8 kDa composed of 16 identical 58.8 kDa. The recombinant thermosome complex was purified in a three-step procedure, including heat treatment, size exclusion chromatography, ion exchange chromatography. The final preparation contained only minor amounts of nucleotides and was 95 % pure from other polypeptides.

ATPase activity was determined for several conditions and the highest hydrolysis rate was found at a pH of 5.7, 1.5 M KCl, and 60 °C. The chaperone function was investigated using the non-native mesophilic model protein citrate synthase. The results showed that the recombinant M. janaschii thermosome successfully prevents thermal aggregation at 45 °C and aggregation upon renaturation of chemically denatured at 25 °C of the citrate synthase at a 1:1 molar ration. Initial reactivation assays of chemically denatured citrate synthase indicated that no release of the substrate was achieved at 25 °C and no regain of activity was detectable.
